• Misako – Voiced by Kayli Mills, Misako is a hot-headed girl, impulsive and ready for a fight all the time. She is Kunio’s girlfriend and a Sophomore Senior at Nekketsu High School.

• Kyōko – She is yet another playable character who is quite the opposite of Misako. Voiced by Kira Buckland, Kyōko is a cheerful, cute, and bubbly girl. She is Riki’s girlfriend and a friend of Misako.

• Kunio – He is one of the main protagonists of the River City Girls and Kunio-kun franchise. Voiced by Greg Chun, Kunio is a kind-hearted yet hot-blooded guy who acts tough when it’s needed. As for his love life, as mentioned earlier, Misako is Kunio’s girlfriend. 

• Riki – He may look like a delinquent, thanks to his hairstyle and dressing sense, but Riki is a much calmer and stoic person in actuality than Kunio. He serves as Kunio’s rival and best friend in the game. Riki is voiced by Kaiji Tang, and he is the boyfriend of Kyōko.

• Marian – The main heroine from the Double Dragon franchise, Marian is a damsel in distress, finally getting her debut as a playable character for the first time with River City Girls 2. 

• Provie – A friend of an NPC named Chris, Provie is a playable character that joins the River City Girls 2 team in search of Chris, who she thinks got kidnapped by the Sanwakai gang, the antagonist group.
